Cagrilintide is a long-acting amylin analogue developed by Novo Nordisk for metabolic research and as a component of combination therapies. Amylin (Islet Amyloid Polypeptide, IAPP) is a peptide co-secreted with insulin from pancreatic beta cells that plays complementary roles to insulin in glucose homeostasis — slowing gastric emptying, suppressing glucagon, and producing satiety signals through CNS amylin receptors.
As an amylin analogue with extended half-life, Cagrilintide targets amylin receptors (combinations of RAMP1/2/3 with the calcitonin receptor) in the brain — particularly in the area postrema and nucleus of the solitary tract — producing appetite suppression through mechanisms distinct from GLP-1 receptor agonists. This complementary mechanism has positioned amylin analogues as combination partners for GLP-1 based therapies.
