MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino acid peptide encoded within the mitochondrial genome — specifically within the 12S rRNA gene. Discovered by Changhan David Lee's lab at USC in 2015, MOTS-c represents a novel class of biologically active peptides called mitochondrial-derived peptides (MDPs), which act as retrograde signals from mitochondria to the nucleus and other tissues.
MOTS-c is sometimes described as a "mitochondrial hormone" — it is secreted by mitochondria in response to metabolic stress and travels to the nucleus to regulate gene expression, and has also been detected in the circulation at concentrations that suggest endocrine-like systemic signaling. Plasma MOTS-c levels decline with age, and exercise increases its expression — properties consistent with its proposed role in metabolic homeostasis and longevity.
