Melanotan II (MT-2) is a cyclic synthetic analog of alpha-melanocyte-stimulating hormone (alpha-MSH), engineered for enhanced potency and resistance to enzymatic degradation compared to the linear native peptide. It acts as a broad melanocortin receptor agonist with affinity for MC1R, MC3R, MC4R, and MC5R — a wider receptor engagement profile than PT-141 (Bremelanotide), which has reduced MC1R activity.
MT-2's MC1R agonism produces melanin synthesis in melanocytes — increasing skin pigmentation in a manner similar to UV exposure but without DNA damage. This tanning effect has made MT-2 a distinct research tool for studying the melanocortin signaling axis and melanocyte biology. Its MC4R activity, shared with PT-141, engages central nervous system pathways involved in appetite, metabolism, and physiological function.
MT-2 is an important research compound for understanding the full melanocortin receptor family and has contributed substantially to the pharmacology that led to the development of approved melanocortin drugs including PT-141/Bremelanotide.
