Semax is a heptapeptide derived from the ACTH (adrenocorticotropic hormone) fragment 4–7 (Met-Glu-His-Phe), combined with the sequence Pro-Gly-Pro to extend its stability. Originally developed at the Institute of Molecular Genetics in Moscow in the late 1980s, Semax has been studied for decades in Russia as a neurological and cognitive-enhancing research compound, and has achieved pharmaceutical approval in Russia and Ukraine for neurological indications.
Semax's primary mechanism involves upregulation of brain-derived neurotrophic factor (BDNF) and its receptor TrkB, as well as stimulation of the dopaminergic and serotonergic systems. This contrasts with Selank (its "sister" nootropic peptide), which produces anxiolytic effects via GABAergic modulation — the two peptides represent opposite ends of the nootropic activation spectrum.
Semax is typically described as a cognitive stimulant and neuroprotective agent, with documented activity in ischemic stroke models, attention and memory research, and stress-resistance studies.
